The A Star Is Born actor and director has received an offer for the role, with both sides engaging in talks, according to Variety.
Leonardo, 44, is said to have pulled out of the movie, which will be based on William Lindsay Greshams novel, after not being able to reach a deal, with Bradley set to take his place.
A movie of the same name, produced back in 1947, saw Tyrone Power star as a con-man who teams up with a female psychiatrist.
However, things start getting sour when the psychiatrist, played by Joan Blondell, starts manipulating him.
The drama is co-written by Del Toro and Kim Morgan, and is set to start filming this autumn.
Bradley, 44, has had a rocky time of it recently, after his split from his girlfriend of four years, Irina Shayk.

More recently, it was claimed that the model accused her boyfriend of having an affair with A Star Is Born co-star Lady Gaga, and was left unimpressed by their friendship.
Bradleys relationship with baby mama Irina began to really fall apart when he started filming with Gaga and she accused him numerous times of cheating on her with his co-star, an insider said.
Ever since then, Irina has not been able to trust Bradley at all.
The source suggested the former couple would fight whenever they were together, with the 33-year-old threatened by their professional relationship.
They added to Radar Online that she found it inappropriate that he started spending time with Gaga at odd hours.
Bradley and Irina, who share their daughter Lea, went public with their relationship back in 2015.
After Gaga and Bradley sparked rumours with their intimate performance of Shallow at the Academy Awards, the Born This Way songstress denied any romance between the pair of them.
